Filtering and Sterilizing

How do you guys filter and sterilize?

I don't know if I'm just over complicating things but I plan on using .45 and then sterilize again with .22 filter.
 
Heat your shit up then push it through the filter into your vials....do you have your vials and beakers and flasks sterilized? That's a very vague question. What specifically are you confused about?
 
Theres a long and decent process I used to use bro:
Clean all areas where you plan on doing this with bleach wipes/ or something similar.
Clothes all windows and any source of a draft to blow dust or particles all over
Sterilize your equipment/vials/ funnels/ hands etc
Heat your oil in the vial and set it in a warm pan of water before you filter it
I used a .22 almost all the time/takes forever but patience is something to always remember
Heres what I do- 18 gauge to draw up the premixed warmed solution I have the clean vial im filtering into ready with the 18 gauge and a 23-25 guage in it to vent it. Draw up the oil in the syringe, turn it upside down cam the 18 guage and unscrew it then screw the syringe onto the filter and needle that's in the sterile vial and push slowly and constant. Then repeat, repeat repeat..I do 20 mls with one filter, then replace.

I read a good whatman .22 can do 100ml-150 on a good day if you don't kill it with super hot oil. Personally I've done...around 120ml. But wow that takes a while. I think that took me almost 2 hours. But that little whatman filtered all that shit.

Sterilizing the equipment isn't the confusion. I read everyone's post about filtering and sterilzing the gear but using a .45 filter. When I took chemistry in HS of all the crap I retained was running it through a .22 filter or stericup and then it is considered filtered or sterile. I assume .22 or .20 is too small at first so maybe a .45 then and then smaller?
 
Area prep is very important. I use a combination of bleach and alcohol and usually clean through out the process. I use my oven to heat everything prior to during and after the process. For filtering I use the Milipor vaccume pump with a hand held brake bleeder.

With that logic I have to ask, what are you filtering?
If a .22 wont work at first then you must have particles that are large or debris.
I completely understand your point and handling excessively could also cause more risk to the solution of infection and a weaker solution and less ml/ml.
So I will let someone else chime in here..I would go straight for the .22 if I didn't have any debris or PG.
 
Guys do it through sterile .45um filters. You can do a one time filter through the .22. Might be a little slower that's all. My last batch I just used a sterile .22 once through. Actually I forget why the .45 is used at all....yeah man it is a SLOW process. Put on some music or something and make sure you are not disturbed.

Just test. I ordered my supplies and well I'm OCD as hell and like writing everything out and planing it. I'll go straight to the .22 and see how hard that is.
 
I would say 40ml will take roughly 50-60 minutes to filter. Unless you have a contraption like a calk gun to make it a little easier. And I would use like a 10ml syringe or something with obviously an 18g pinn. Anything bigger than 10ml will just be harder.
 
If you don't have a large syringe only do half of any others. That little plunger will feel like a knife going through your thumb with a .22 and the constant pressure and the time consumption. I always did mine in the evening when it was calm, no disturbances and no chance of anyone bothering me.
 
I forgot if you are using sealed and sterile vials put a smaller pinn through the top for air to escape as you fill or else pressure will build up in the vial and it will be almost impossible towards the last few mls. It was easier for me to use open vials that I sterilized myself. I just attached the filter to the end of the syringe and just filled open like that then put the little stopper in as soon as I was done. Make sure all fans are off so dust is minimized. But that is what the higher ba % is for. For us guys a little more worried about sterility and germs homebrewing just up the ba%.

I was curious; how secure is the stopper without crimping it? I have vials without tops from a previous order that was filled incorrect and was curious if the stopper is pretty snug on there
 
if its crimped with a metal band its pretty secure. If not with pressure it can pop off. They fit good but if your putting sterile liquid in it then vent it..however im pretty sure you can get sterile sealed vials..without giving to much info openly..use google.

yeah, med lab supplies is where I get my vials but they sent me about 20 open vials by mistake. Thought I could finally put these for use lol
